Design of SARS-CoV-2 hFc-Conjugated Receptor-Binding Domain mRNA Vaccine Delivered <i>via</i> Lipid Nanoparticles

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been identified as the causal agent of COVID-19 and stands at center current global human pandemic, with death toll exceeding one million. The urgent need for a vaccine led to development various immunization approaches. mRNA vaccines represent cell-free, simple, rapid platform immunization, therefore have employed in recent studies toward SARS-CoV-2 vaccine. Herein, we present design an vaccine, based on lipid nanoparticles (LNPs)-encapsulated Fc-conjugated receptor-binding domain (RBD-hFc). Several ionizable lipids evaluated vivo luciferase (luc) reporter assay, two leading LNPs formulations chosen subsequent RBD-hFc strategy. Intramuscular administration LNP elicited robust humoral response, high level neutralizing antibodies Th1-biased cellular response BALB/c mice. data study demonstrate potential these promising candidates LNP-based general COVID19 particular.
